I am really confused on skills needed to do a job. How do I know what I need? I have the skills that are needed for the job but I still cant do it.

Each job has 5 skills you need (maybe one will be repeated two or three times) and a certain number of labour points - so the total of your skills (with item bonuses) needs to be greater than the required labour points. If you meet that requirement, you should be able to do the job.

the skilled needed has to added up greater to the labour points or the difficulty?

Your labor points need to be higher than the difficulty.
