No you do not have to join a town to do ANY of the quests.
For that particular quest, you need to do the job 'Selling Newspapers' until you have 3.
Then the next quest is 'The Mean Henchmen' and you need to do the duel (just click 'Challenge Rival' or 'Complete' on the quest page until you win.)

It pains me sometimes, it really does. It is hard to stay patient.
To get to that quest, you have to do others. It makes me wonder how they got that far. And I wonder if I am really helping, or just prolonging the misery for them as I doubt people like our friend sheik here are going to make it to the high levels.
