I've been spending the past few days trying to figure out whats going on but I can't seem to get the Day of the Dead quest line. No matter what I've done it doesn't come available for me.

You have to finish the quest Showdown (Save the saloon, part 2). Once you do that, the first quest for Day of the Dead should be available in the Saloon from Maya Roalstad.
Also keep in mind that you have to accept the quest between October 27, 2017 and November 14, 2017. So its too late at this point to start it.
